Research Focus
Our main research interest is to understand
the cellular responses to cerebral ischemia with the goal of developing
useful anti-ischemic therapies. While much of the work is carried out in
primary culture to analyze behavior of isolated cell populations, we also
use animal stroke models and acutely isolated preparations to study the
cellular responses to ischemia, and ways to modulate them. In addition
to changes in gene expression with injury we are interested in the interaction
of neurons and astrocytes during injury, and defining the differences in
vulnerability between these two cell types.
Gene therapy of stroke
We are currently using transgenic mice and retroviral expression in
primary culture to study the effects of changes in gene expression on cell
survival from ischemia and ischemia-like injuries. Genes currently under
study are hsp70, bcl-2, bcl-x, E6, E7, SOD1 and 2.
Effects of acidosis during ischemia, regulation of pH in ischemia
We have cloned a splice variant of the sodium bicarbonate cotransporter that is expressed in glial cells in the brain. We are currently studying its physiological role and the effect of altering its expression on outcome from ischemia.
